The Screening of a High-efficiency organophosphorus pesticides -degrading strain and its Enzyme Properties

Abstract: Abstract: Objective: The test is to screen and identify a high-efficiency biodegradating strains from the contaminated soil to a high concentration of organophosphorus pesticides and its enzymology properties were studyed. Methods:In this study, A high-efficiency organophosphorus-degrading strain was selected through domestication, separation, screening process and identified by a series of colony morphological, physiological characteristics and 16s rDNA sequences and other tests. And the crude enzyme liquid enzymatic properties were studied by the step of pH, temperature of the enzyme, metal ions and etc. Results:A strain J7-4 was screened and identified as Pseudomonas stutzeri. The results showed that the degradation enzyme is located in the cell .The optional reactive pH and temperature of the enzyme is 45℃and pH9.0;The most suitable substrate of the enzyme is methyl parathion, and it has a broad-spectrum degradation characteristics; Some metal ions such as Mg2+, Ca2+ had activation on the enzyme at different degree, while Co2+、Cr3+ have slight inhibition on the enzyme, and Mn2+, Fe3+, Zn2+, Al3+, Hg+, Cd2+, Ba2+, Ag+ have a strong inhibition on the enzyme. Conclusion: A strain Pseudomonas stutzeri J7-4 was screened successfully with high efficient degradating Organophosphate compounds and its enzymology properties were studied.
