Identification of genetic relationship of corn germplasms with molecular markers and evaluation of their salt tolerance

Abstract: In order to analyze the genotypes of 10 corn accessions, detect their salt-tolerant level and reveal salt-tolerance machanism in corn, 20 pairs of simple sequence repeat (SSR) molecular markers were used to amplify the 10 corn accessions, and the corresponding SSR fingerprints were obtained to collect 0, 1 data. The method of NJ cluster analysis was performed to construct dendrograms. Meanwhile, NaCl solution with the concentration of 0, 100, 200 and 300 mmol/L, respectively was prepared and employed on the corn seedlings. Plant height of the corn materials was measured as the reference data of salt resistance. After seven times of reatment with NaCl solution, the activity of antioxidative enzymes including superoxide dismutase (SOD) and peroxidase (POD) was detected. The genetic relationships of the 10 corn accessions were identified according to the molecular marker data, with the genetic similarity coefficient varying from 0.54 to 0.71. Based on the plant height data and the results of enzymatic activity that 5 salt-resistant corn materials were chosen, which were Xianyu335, Zhengdan958, LH214, 83IBI4 and LH204. The results will be helpful to choose the best parents for corn salt-tolerant breeding.
