The Singleness Analysis of Monokaryon from Agrocybe aegerita

Abstract: To study the singleness of monokayon from basidiomycetes, the morphology and molecular identification were compared for providing a reference for hybrid breeding and genetic research on edible fungi. The single-spore strain was isolated using the spore-dilution method, and then the clamp connection structure was observed. The decidable “monokayon” was further analyzed based on the amplification of A and B mating loci. The results showed: (1) Of 11 single-spore strains, 10 strains were identified for the monokaryon except 1 dikaryon producing clamp connection structure; (2) The amplified results based on the A and B mating loci showed 3 non-singleness “monokayons” contained two A and one B mating loci. The “monokaryotic strain” which identified by the clamp connection was probably mixed with heterogenous strain carrying the identical A or B mating loci, and it was suggested for obtaining monokaryotic strain using the single-spore isolation method combined with molecular analysis.
