Core Primer Screening on Cultivar Identification of Beet with SRAP Molecular Marker

Abstract: Using 12 foreign beet varieties as materials, 546 pairs of SRAP primer combinations were amplified and then separated with gel electrophoresis of 8% non-denaturing polyacrylamide. The SRAP core primer combination which was the best for the purity identification of beet varieties was selected. As a result, 23 pairs of SRAP primer combinations with clear amplified bands, rich polymorphism and good repeatability were screened out from the 546 pairs. And the 23 pairs were amplified into 177 polymorphic bands. The PIC values of the primers were all greater than 0.6. The number of polymorphic fragments was 7.7 per primer pair. In theory, these core primers can satisfy the need of the identification of the existing beet varieties.
