Gene Expression of Aphelenchoides besseyi trehalase (Ab-tre) Gene under Hypertonic Osmotic Stress

Abstract: Trehalose can be hydrolyzed by trehalase in rice white tip nematode (Aphelenchoides besseyi ), which provides energy to enable the survival under hypertonic osmotic stress. This study aims at studying the gene expression of A. besseyi trehalase (Ab- tre ) gene. Ab- tre was cloned based on the transcriptome sequencing result. A 30% glycerol solution was selected to be the dehydrant to provide a hypertonic osmotic stress and the expression ofAb-tre under the hypertonic osmotic stress was studied by Q-PCR and in situ hybridization. Q- PCR showed that the gene expression of Ab- tre increased significantly when the nematodes were undergoing hypertonic osmotic stress for 3-7 min or recovered in distilled water for 90 min after a 140 min treatment of hypertonic osmotic stress. In situ hybridization showed thatAb-tre was mainly located in the central of the nematode. This study indicated that Ab-tre was related to hypertonic osmotic stress resistance ofA. besseyi . The study on the function ofAb-tre can provide a theoretical basis for the prevention and control of the nematode.
