小麦GAPDH 基因克隆及序列分析

Cloning and Sequencing of GAPDH Gene in Common Wheat(Tritucum aestivum L.)

摘要: 采用Trizol法提取高质量小麦总RNA,并运用RT-PCR方法克隆了小麦GAPDH (甘油醛三磷酸脱氢酶)基因的部分序列(EU022331),长度为604 bp,核苷酸序列比对结果表明,该序列与大麦(M36650)、玉米(AY109397)、番茄(BT012693)GAPDH核苷酸序列的同源性分别为95%、86%和84%。GAPDH基因的克隆为在禾谷类作物研究的应用提供了一个较好的对照基因。

Abstract: The total RNA of wheat leaves was extracted with Trizol, and the partial cDNA sequence of wheat GAPDH gene(EU022331) was amplified through RT-PCR. The colned GAPDH gene (604 bp) was 95 %, 86 % and 84% identified with to M36650(Barley), AY109397 (maize), BT012693(tomato) , respectively. This partial sequence provided a good background on studying expression of many cereal genes.
