The Morphology Observation of Infection Process for the Pathogen Exobasidium vexans of Tea Blister Blight against Tea Leaf

Abstract: To determine the morphological characteristics during infection process for the pathogen of Tea Blister Blight, this should facilitate the disease diagnosis and effective control. This study used the stains of Periodic Acid-Schiff and Toluidine Blue O to analyse the infection process of the pathogen against the tissues and cells in tea leaf. Meanwhile, Trypan blue and Evans blue were used to analyze the cell viability of the tea leaf, and the status of basidiospore and hypha. The histological stain and TEM found that an amount of spore germinated and hypha grown during 2-3th phase, and the nutrient of tea leaf cells was gradually consumed, then the viabilities of the basidiospore and hypha were also decreased, tea leaf cells ultimately tended to die. This study found the morphological characteristics during various infection processes by the methods of the histological stain, and found the influence of cells from the spongy tissue triggered by the pathogen.
