Construction of Chicken IL-2 Recombinant Baculovirus and Its Histopathological Study

Abstract: To investigate the effect of chicken interleukin-2 (IL-2) recombinant baculovirus (BV-LMI-IL-2) on chicken visceral organs and provide scientific basis for developing newcastle disease (ND) vaccines, IL-2 was taken as the immune stimulating factor. The gene encoding IL-2 was inserted to the baculovirus transfer vector pLMI, thus BV-LMI-IL-2 was obtained. Finally BV-LMI-IL-2 and ND antigen vaccine BV-LMI-F were used to conduct independent and combined immunization. At the same time, pathological changes in organs including glandular stomach, liver, heart and duodenum were observed. Immune and challenge experiment showed that the protection rate of combined immunization of BV-LMI-IL- 2 with BV-LMI-F increased by 12.5%. This indicated that BV-LMI-IL-2 could enhance the immune function and be used as an immune enhancer. The chicken in the control group and immune group were dissected. Observation of diseased organs and histopathologic slide showed that organs in combined immunization group were without obvious pathological changes. It was proved that the chicken IL- 2 recombinant baculovirus improved the immune protective effect on chicken organs.
