  Study on RAPD Markers of Cashmere Yield and Body Weight of Inner Mongolian Cashmere Goat Zhang Enping, Geng Sheming, Zhang Yani, Jia Zhihai 2004, 20 (4):  1-1.  Abstract + ( 1171 )   PDF ( 582356KB )   ( 576 ) 12 random primers were used to pre-amplify genome DNA of 27 inner Mongolian Cashmere goat, and 4 primers which producing better polymorphic DNA were selected on RAPD analysis. 51markers including 47 variable markers were observed in this study. The polymorphic frequency was 0.84 and the length of amplified fragments among176~2940bp. Studies of relationship between RAPD markers and cashmere yield and body weight of Inner Mongolian cashmere goat showed that: ABCFHO of CY0816 and INR of F09 were superior combined types on cashmere yield; GIJOPQR of CY0816was superior combined type on body weight.

It was at the first time found that the main chemicals in T. kirilowii are alkaloids and flavone, that the extraction prepared with 60 percent of ethanol solution performed the best effects on prolonging the life span of used mice.

The results showed that the number 7 was the best extender (p<0.05)in seven groups, and the best cryoprotectant was glycerol (p<0.01).When the glycerol content was 2 percent (p<0.05),the sperm motility of frozen semen pellets was best in all treatment groups.The method of dry thawn of frozen semen pellets was better than that of wet thawn(p<0.05); the sperm motility was 0.46-0.49 after the frozen semen pellet was thawed at 40 to 45 ℃. supplyment in the extender could increase the thawn sperm survival time and motility effectively(p<0.01).

The straws were rozen with programⅠand programⅡin the programmable freezer separately. The mean post thaw progressive motility （PTPM）were 0.47±0.10 and 0.34±0.05 (P＜0.01) resp., and the percentage of sperms with intact acrosome were 34.47±5.20 and 29.77±4.37（P＜0.01)resp. It is concluded that the programⅠwas better than program Ⅱfor freezing canine straw semen.

A high oocyte maturation, fertilization and cleavage rate and a low rate of blastocyst yield and calving following transfer of in vitro produced buffalo embryos have been obtained. The efficiency of IVP in buffalo is much lower than that in cattle. Several problems need to be resolved before IVP technology can be used regularly in buffalo breeding. This review attempts to present an overview of the different techniques used in buffalo to produce transferable embryos in vitro, namely in vitro maturation and fertilization of immature oocytes and in vitro development of the resulting cleaved embryos to the blastocyst stage before transfer. The problems associated with IVP and the possible solutions discussed.

  Purification and toxicity characterization of Ricin from Ricinus communis beans Zeng You-wei, Peng Yong-hong, Song Guang-quan　Xu jie, liang guan-sheng 2004, 20 (4):  23-23.  Abstract + ( 1888 )   PDF ( 751416KB )   ( 855 ) Ricin was purified from Ricinus communis beans to apparent homogeneity by ammonium sulfate precipitation, Sepharose-4B affinity chromatography, and followed by molecular sieve chromatography. The purified preparation showed two protein bands with relative molecular weights of 32 kDa and 34kDa when subjected to sodium dodecyl sulfate-polyacrylamide gel eloctrophoresis. The isoelectric point of the purified Ricin was pI6.1 and pI6.7 determined by isoelectric focusing (IEF). There were five peaks when Ricin was subjected to high performance liquid chromatography (HPLC) under non-denaturing conditions, indicating this Ricin consisted of five different multimers. Purified Ricin revealed high toxicity to mouse, but had no any toxicity to Prodenia litura Fabricius larvae.

  Evaluation of Some Characters of Insect resistant Cotton on the Cotton Productivity Wu Zhengbin, Yang Yehua, Zhang hong, Zhu Renshan, Chen Jinshu and Chen Guoping 2004, 20 (4):  26-26.  Abstract + ( 1539 )   PDF ( 824145KB )   ( 658 ) Two classes of insect resistant varieties and four groups of traditionally bred varieties were investigated in their economic and agronomic characters, early maturity and pink bollworm resistibility in the years of 2002 and 2003. The tests were arranged in accordance to the design of randomly grouped plots and three times repeats. The identification of pink bollworm resistance was carried out in caged plots. The results showed that the insect resistant varieties, especially the insect resistant hybrids, expressed a better cotton productivity and produced a higher yield under the condition of 70% of reduced chemical prevention and control of the pests compared with those of the ordinary cotton growing field. The percentages of the yield increments reached extreme significant levels statistically. A conclusion was made that breeding out and popularizing the insect resistant cotton varieties had a broader future, as these varieties had a higher fiber quality, stronger insect resistibility and earlier maturation character.

And the average devirus rate was 83.7% based on the semiological diagnostic and plant indicator identification. The reproduction coefficient for the fast propagation with cut test-tube plantlets was 5 n, the reproduction speed increased at a geometric progression and large-scale seedlings could be multiplied in a short time. The virus-free seedlings transplanted in fields reached a yield increase of 37.8%.

Treatment with different concentration of 6-BA was attempted for adventitious shoot induction. Lower concentration（1.5 mg/L） of BA was suitable. MS+BA2.5 mg/L+NAA0.05~0.1mg/L was better for buds proliferation with the proliferation coefficient 4.1every month. Seedling transferred in 1/2MS medium with 0.2~0.5mg/L NAA could root normally.

  Efficient Methods for Genomic DNA Extraction from Actinidia Deliciosa Xu Xiaobiao， ，Chen Hua，Zhang Qiuming 2004, 20 (4):  41-41.  Abstract + ( 1589 )   PDF ( 733876KB )   ( 798 ) In order to isolate high quality DNA from Actinidia leaves enriching polysaccharides and polyphenols and preserved from field samples, the effects of DNA extraction were studied with four different methods including CTAB regular method, CTAB subarea method, SDS regular method, SDS subarea method and three different conditions of storing sample (frozen at -20℃ and -70℃, dried with silica gel). Moreover, the CTAB subarea method was used for the first time for genomic DNA isolation from Actinidia .The results showed that the CTAB subarea method and dried with silica gel was optimal, the value of OD260/OD280 was over 1.8,the DNAs were completely digested by restriction endonuclease and the extracted DNA by this method was appeared as a clear band when electrophoresed in 1% agrarose gel, and produced clear polymorphic patterns amplified by PCR .

The new criterion for the optimum conditions was put forward , that the optimum conditions was the incubation temperature and the pH when the enzyme inhibition rate was maximum, that the precondition was enzyme was on certain activation level（0.1333 min-1 ≤k1 ≤0.2667min-1 [ 1 ]）.The optimum incubation temperature was 47℃and the optimum pH of phosphate buffer was 7.7 when 10 minutes was chosen as the incubation time. The new criterion of the detection result in the optimum conditions was put forward ,that vegetable contained certain organophosphate or carbamate pesticide residues, if enzyme inhibition rate≥58%.

The result indicated that with proper handing to treatment mushrooms could significantly reduce the cap opening and quality deterioration during storage. The experimented results was further analogged by using regression equation. The estimated values were found to be identical with the measured values. Compared with regular low-temperature storage, the mushrooms had better quality and physical-chemical index in low-temperature and controlled atmosphere storage.

In the organic tea garden, the population of Empoasca(Emposaca)vitis(Gōthe)was lower but the population of spiders was higher compared with the conventional tea garden. The correlation coefficient of the population dynamics between Empoasca(Emposaca)vitis(Gōthe)and spiders in organic tea garden(r：0.2182~0.7355)was higher than in conventional tea garden(r：0.1318),so organic management can help the interacting relation between pests and natural enemies and come to being.

  Study of brewing experiments on new grape varieties of South China Peng Hongxiang, Zhu Jianhua, Huang Honghui, Lin Mingjuan, Huang Fengzhu 2004, 20 (4):  82-82.  Abstract + ( 1273 )   PDF ( 675905KB )   ( 684 ) Fruit and vintage characters of mature berry of 6 new selected grape varieties (serial number) of South China were tested with evaluation for further brewing experiments. All varieties were tested with brewing technique of traditional red wine except that “V-32” was done with technique of dry white wine. The results showed that soluble solids and total acid content of Nanbaoshi and Nanzhenzhu were relatively high with 17.0%、12.74 g/L and 19.0%、10.3 g/L respectively. Juicing output of “V-64” was the highest of all, up to 73.3%, while total sugar, reductive sugar and tannic substance of “V-63” were relative low. Sense observation of vintage samples showed that“V-32”was suitable for dry white wine brewing, “V-64” and Nanbaoshi and Nanzhenzhu for sweet wild grape wine. And Nanbaoshi was of high commercial potential for suitable for fine dry wild grape wine brewing. “V-59” was good for fresh consume, and “V-63” was not ready for grape wine production.

  Study on the interaction among soil additive, soil microorganism and tomato seedling stage Qu Zaihong，Du Xiangge 2004, 20 (4):  84-84.  Abstract + ( 1313 )   PDF ( 687854KB )   ( 748 ) Based on the characteristics of growth of plant organism, add organic additive made of the juice of alfalfas, wooden vinegar liquid, spring onions and juice, 4 kinds of plant ash and organic manure into the soil according to the growth stage, setting organic manure treatment as control. The result of this study shows: after treatment with this organic additive, the quantity of the soil microorganism increases markedly, meanwhile, the height &weight of tomato also have obvious changes, of which the changes of treatment group B is most obvious. Compared with control, the bacterium's quantity in the soil increases by 65.88%, 96.14%, 80.90% and 82.0% in 15d, 30d, 45d and 60d respectively; The quantity of soil antinomies increases by 16.01%, 54.01 %, 49.0%, and 49.8% respectively in the same time points. Accordingly, the average of height&weight of treatment group B increases by 35.22% and 34.97% respectively during the last period of seedling stage of the tomato. ANOVA analysis shows strong positive correlation between bacterium&actinomyces quantity and the height&weight of tomato seedling in various times of measurement.

Researches on drought resistance mechanism have been being a hot spot for many years, and various kinds of morphological and physiological indexes for drought resistance have been put forward, but so far, the study on drought resistance mechanism is not in- depth, and the indexes put forward are not perfect in practice. This paper showed the views for drought resistance mechanism in plant.

It was showed the Dongzao jujube cultivar had an optimum pH range，the value is 7.0. The optimum native substrate of PPO in ‘Dongzao’ was gallate.

  The Study of Factors of Effecting on High Frequency Shoot Regeneration From in vitro Leaves in Pear Sun QingRong, Liu QingZhong, Zhao HongJun, Liu Peng 2004, 20 (4):  99-99.  Abstract + ( 1439 )   PDF ( 419856KB )   ( 546 ) The factors of effecting on shoot regeneration from leaves in pear were studied. These factors included genotype, basic media, plant growth regulators, ways of leaf inoculation and culture condition of dark and light. Genotype is the most important factor of effecting on shoot regeneration from leaves. The highest rate of shoot regeneration were reached from D’Anjou and Hosui, both reached 100%，the lowest is Yellow Gold pear, only 27.3%. Plant growth regulators needed for different genotypes reaching the highest regeneration rate were different. TDZ is more effective than BA for inducing shoot regeneration from Duckpear, and BA is more effective than TDZ for inducing shoot regeneration from Pyrus communis L. and P. pyrifolia Nak. Shoot regeneration is improved by culturing 3 weeks under dark. The ways of leaf inoculation on media had less effect on shoot regeneration.

It shows good quality characters, such as longer development time and longer stability time. Useful conclusion can be drawn from its successful breeding: It is possible to combine good quality characters with high kernel yield potential; Two parents should be complementary each other on main quality characters and be multiple on its genetic background; The breeding of good quality wheat variety must rely on essential and valid quality characters testing means.

  Studies on Flowering Habits of 5 Newly-developed Japonica Photoperiod/ Thermo-sensitive Genic Male Sterile Lines He Xiaopeng, Yang Shuhua Tian Yawen Li Xuelin 2004, 20 (4):  104-104.  Abstract + ( 1557 )   PDF ( 593663KB )   ( 640 ) The flowering habits and outcrossing characteristics of 5 newly-developed japonica photoperiod/ thermo-sensitive genic male sterile lines were studied. Results showed: The flowering duration of single plant and single panicle of Y27S is the shortest, 118S is close to control N5088S, B30S, 116S and 117S are comparatively longer. Flowering kinesis of every day in a panicle of Y27S, 116S, 118S and 117S are similar to that of restorer line 41678, but B30S lag behind 41678, control N5088S is earlier than it. Flowering kinesis in a day in a panicle of Y27S, B30S and control N5088S are close to that of 41678. Although the anthesis time of 116S is dispersed, its flowering is earlier and forenoon flowering rate is relatively higher. The anthesis time of 117S is concentrated and the flourishing flowering is late, while 118S is dispersed and late. Spikelet opening time of 116S and 118S is the longest, 117S is relatively short. As for the stigma exsertion rate, 118S is the highest, 116S is the lowest, and that of 118S, B30S and 117S are more than 60%. The lasting time of the stigma vitality of Y27S is the longest. The fall of the stigma vitality of control N5088S is the slowest. All the P(T)GMS have higher wrapround spikelet rate and relatively lower indehiscent spikelet rate.

The results showed that (1) plant height, internode length No.1, panicle length and pikelets per panicle show the characteristic of quantitative traits. (2) There is the clear difference on correlation of some traits between indica and japonica rice regions, which shows plant height and seed setting rate etc. traits at cold-cool jaopnica rice cropping regions. On the contrary, panicle exsertion and four trait of panicle of RIL were significantly correlated at 5% or 1% level of at high temperature indica region. (3) Genetic analysis showed that there were major gene affecting the panicle exsertion and seed setting rate of low or high temperature sensitive, a lot of weaker plants with cold tolerance have relatively stronger heat tolerance.

Based on the difference of cotton fiber quality in three regions, E-dong, E-bei, and Jianghan Plain should be built to produce three main types cotton, respectively. Huining-18 grows semi-stolon seedlings and deep-colored leaves; it has spindly white ear, long awns and red grains. the mass of 1000 grains ranges from 30.5 to 46.3g, protein content is 18.74%. Its adult plant is immune to prevalent stripe rust such as Tiaozhong 31 and Tiaozhong 32. The cultivar also shows good drought tolerance.

The available Al、P、Ca content and distribution in root region showed difference between two kinds of soil with different pH, but the content and distribution of available K was basically same. The Al content in the acid soil near the root (0-1cm) was significantly higher than that in far-from-root region. The Al content in soil of root region of Al-tolerant inbred lines Z01 was significant higher than that of Al-sensitive ones, and Al-tolerant inbred lines Z01 had strong excluding capability to Aluminum. There was a P gradient in 0-4 cm region that declined from far-from-root region to root surface in two inbred lines in neutral soil, but the P declined gradient only exist in 0-2.5 cm region near root surface in acid soil. In acid soil, the P content of Al-tolerant inbred lines Z01 in 1-2.5 cm region near root surface was relative higher and mobilizing ability to P was relative stronger. The Ca content of root region in the acid soil was significant lower than that of neutral soil. In acid soil, the Ca、Mg absorbing capability of the roots in Al-tolerant inbred lines Z01 was relatively stronger, and an obvious Ca declined gradient in 0-2.5 cm region from far-root region to root surface was found in tolerant line, but in sensitive lines Z02 it had a small change.

In order to develop Talinum paniculatum fully, it was necessary to strengthen the research on genetic improvement to invent a good variety. A new technique of genetic improvement was supplied in this paper, which was called ion beam biotechnology.

The results indicated that the light and temperature in winter-spring was more beneficial to cucumber growth and development than in autumn-winter. In winter-spring cucumber root volume increased more quickly and the vitality of root increased gradually, autumn-winter root volume increase slowly and the vitality of root gradually descended. Uptake amount and intensity of Boron of cucumber in winter-spring was also obviously higher than in autumn-winter. Distribution of Boron in different part of cucumber in winter-spring was higher than in autumn-winter, with dominant tendency to leaf and lack to root. Application of Boron should adapt to the environment and plant development stage in practice.

Plantlets of rooting-stage could survive on the sucrose-free media .which showed the characteristic of photoautotrophical growth , plantlets grown in the vessel closed with the gas-permeable film closure have greater survival rate , fresh weight ,dry matter and rooting rate than those closed with airtight film. gas-permeable film closure also increase the proliferation rate of explants. The result showed the potential of controlling culture enviroment to improve the efficiency of micropropagation and quality of plantlets through photoautotrophical growth.

  Characteristics and Pattern of Change in Nitrogen, Phosphorus and Potassium Content in Various Organs of a Prunus mume at Its Different Growth Stages Sun Qibao， Sun Jun, and Yu Feifei 2004, 20 (4):  232-232.  Abstract + ( 1404 )   The pattern of change in the concentration of nitrogen(N), phosphorus(P) and potassium(K) in organs of three-year-old trees of Prunus mume/wild peach indicates that the change was dynamically corresponded with the annual growth and development cycling of the tree proper. In winter, N, P, and K were stored in all organs of Prunus mume and were translocated to new organs in the following season. N content and offering in thick roots was higher than the other perennial organs, while P, K content and offering of the front was lower than the latter. K was the nutrient element that played the most important role in the process of differentiation of flower-bud. N remarkably promoted the development of young fruit. The three elements concentration in new shoots was higher than in young fruits. It is recommended that the supplementary NP fertilizer should be applied in the autumn and spring, and K fertilizer prior to the flower-bud differentiation period.

  Natural predators of Thrips palmi (Karny) and their role in natural control. The predatory fauna comprised arthropods in 10 families. The anthocorid bugs Orius. Similis Zheng and the mirid bugs Campylomma chinensis Schuh and Dortus chinai Miyamoto were the most numerous and widely distributed species observed, constituting 95% of all predators. Of these indigenous predators, C. chinensis is most likely to be effective against Thrips palmi. 54 % Thrips palmi that were preyed by predators were preyed by C. chinensis. C. chinensis can reproduce all around a year. C. chinensis is one of the most potential enemy of controlling thrips palmi in Southern China. The potential of the main 3 species for biological control of Thrips palmiis is discussed.

In spite of considerable efforts dedicated to soil salinization, the extent of secondary salinization tends to increase. This requires careful monitoring the soil salinity status and variation to curb soil salinization trends, and secure sustainable land use and management. Multi- sensor and multi-temporal and multi-spectral and microwave remote sensing data has been widely used to detecting temporal and spatial changes of soil salinization. Airborne remote sensing and ground-based electromagnetic induction meters and laboratory and field spectrometers, combined with ground soil salinity data, have shown potential for monitoring soil salinization. When the vegetation cover threshold is below 25-35% in semi-arid and arid, soils differ widely in their spectral response, and it is a most effective method to monitoring soil salinity by integrating soil salinity data and soil spectral data. However, if the vegetation cover threshold is beyond 25-35%, the above method doesn’t work well, soil salinity can be only inferred from the above vegetation spectra. The objective of this research is to discuss the relationships between soil salinity and above vegetation field-derived spectra, and use above vegetation field-derived spectra to predict soil salinity status in Liaohe River Delta tentatively. The result shows that it is feasible.
