  Studies on Application Effects of Different Sources of Inactivated Vaccines against PRRS 2009, 25 (5):  6-9.  Abstract + ( 1507 )   PDF ( 1046415KB )   ( 974 ) Abstract: To verify the application effects of inactivated vaccines of porcine reproductive and respiratory syndrome (PRRS) from different factories and to supply experimental evidence for correct selection of different sources of inactivated PRRS vaccines, the most frequently applied inactivated PRRS vaccines from three factories were compared in adverse reactions and immune effect. It was found that the vaccines used did not cause the adverse reactions such as hypersusceptibility and changes of feed intakes, and that 2 times of vaccinations of the vaccines all effectively enhanced the antibody levels, respectively, in which the No. Ⅲ vaccine stimulated antibody as rapid as the first vaccination was done while the No. I and No. Ⅱvaccines did not result in the antibody by the first vaccination. It was concluded that the No. Ⅲ performed the best immune effect in the used vaccines since it was the most rapid one in producing the antibody and can be taken as the first-selection of inactivated vaccine against PRRS in Zhuhai, Guangdong.   Analysis on Antibacterial Activity and Anti-Peptide of Different Growth Stages from Tenebrio Molitor Wang Lixin, Wang Shanshan, Dai Sifa, Lu Zhenxiang, Wang Jinhu, Hu Jing 2009, 25 (5):  10-13.  Abstract + ( 1466 )   PDF ( 1854389KB )   ( 758 ) Abstract: Objective The purpose of the study was to analyze the cecropin antibacterial activity and different component of antibacterial peptide in different growth stages. Material and Methods Tenebrio Molitor produced antimicrobial peptides in different stages, after infected E. coli with the method of acupuncture. Antimicrobial peptides were obtained after grinding and centrifugating the infected Tenebrio Molitor, and determined antimicrobial effects of these antimicrobial peptides with drug-test. Protein and polypeptide were separated with the method of polyacrylamide. Results The results of the sensitivity test showed that bacteriostasis effect of antibacterial peptide of Tenebrio Molitor in the larval stage was more effect than that of antibacterial peptide of Tenebrio Molitor in the pupa and the adult phase, but there was no significant difference. Tenebrio Molitor electrophorogram showed that there were 8 genophore in larvae stage, 5 genophore in prosopon, and 5 genophore in pupa. The result showed that the variety of antimicrobial peptides of Tenebrio Molitor in pupa and adult was less than that of in larvae, however, more little peptide were not separated from. Tenebrio Molito of each stage.

  Development of RT-PCR method for detecting Infectious Bronchitis Virus Cheng Xiaoxia,, Wang Shao,, Zhu Xiaoli, Cheng Shilong,, Lin Fengqiang,, Chen Shaoying, 2009, 25 (5):  14-17.  Abstract + ( 1475 )   PDF ( 772532KB )   ( 851 ) A pair of primers were designed and synthesized based on the sequences of the IBV genomes, and the RT-PCR method to detect the infectious Bronchitis virus （IBV）were developed.The 1105 bp specifical fragment were obtained from the IBV vaccine strain RNA by this RT-PCR.The sensitivity of RT-PCR reached to 2.85 ng IBV-M-cDNA.These results showed that the RT-PCR method to detect IBV were of better specificity and higher sensibility. The results showed that this PCR technique was specific and applied to IBV diagnoses and epidemiology investigation．

Results showed that the methods combination of freezing-thawing and CATB could smash cell wall of varieties of micro-organisms including bacteria and Fungus. DNA extraction was stable by this technique and DNA fragments were large enough to be applied in follow-up research. It could be concluded that freezing-thawing and CATB methods combination could extract the total rumen microbial DNA.

The PCR products were ligated into the pMD-19T vector, and then transformed into competent cells of E.coli DH5a.The positive cloning was sequenced and analyzed.【Result】The cloned nucleotide sequence shared 99.49 % homology with the code zone sequence of mouse prolactin-releasing peptide receptor.【Conclusion】The cloned nucleotide sequence was part of mouse prolactin-releasing peptide receptor gene and prolactin-releasing peptide receptor gene express in mouse different tissues during gestation.

The end that shape of microcapsules was orbicular or elliptic and no conglutination, the best means for testing envelopment protein capacity of microcapsules is the Folin-pHenol assay because it has the least effection.

The effect of them on anther swelling, contamination, browning and embryo production were detected. The results indicated that 0.5~1.0 ml/L Shan Nong Yi Hao Ⅲ was good at controlling anther contamination and embryo production, which could be used for sweet pepper anther culture.

  Study on DNA damages of hemocytes in spider with single cell gel electrophoresis 2009, 25 (5):  48-52.  Abstract + ( 1598 )   PDF ( 2153896KB )   ( 955 ) To study and evaluate genotoxicity of H2O2 and formaldehyde in the spider, pardosa astrigera, the adult of female and male spiders were treated with H2O2 at concentrations of 50,75,100,125,150µM and formaldehyde at 10,25,40,55,70µM, respectively. DNA damage of hemocytes was detected by the comet assay. CASP software was used to analyze the comet pictures. The results show that the percentage of comet, the percentage of tail length, and the tail moment of the H2O2 tested groups were significantly different from those of the control group (P<0.05). A statistically significant dose-dependent increase in DNA damage was observed. Formaldehyde induces DNA strand breaks at 10µM, but crosslinks at 25,40,55µM, and apoptosises at 70µM in hemocytes cells of spiders.

And appropriate conditions of induced mutation can increase mutation efficiency and develop peanut germplasm resources well. Seed explants of two peanut varieties were used and five concentration (0.3%, 0.6%, 0.9%, 1.2%, 1.5%) and five processing time (0.5 h, 1h , 3h, 5h, 7h) were set up. The results showed that the appropriate combinations of zhonghua No.5 were 0.9% 7h and 1.2% 5h and the appropriate combinations of yuanza 9102 were 0.6% 7h and 0.9%7h according to half-death rates for the selection criteria. The obvious physical injury was caused by EMS treatment and the sensitivity of EMS to two varieties were a little difference. Molecular identification results showed that the DNA mutation had already occurred.

The best result for decolorizing hsian-tsao gum was determined as follows: the volumn of resin was 3%, decolorization time was 3h and decolorization temperature was 50℃, under which the decolorizing rate reached 95.5% and the loss of polysaccharide was 15.5%.

  Isolation and Purification of Polysaccharides from Fermented Products of Tremella aurantialba and Hypoglycemic Effect 2009, 25 (5):  63-67.  Abstract + ( 1342 )   PDF ( 1512200KB )   ( 860 ) 【OBJECTIVE】The purpose of this paper is to isolate and purify the polysaccharides from fermented products of Tremella aurantialba (FTAP) and to study its hypoglycemic activities.【METHOD】The process of boiling-water extracting polysaccharides were investigated in comparison with that of alkaline solution extracting polysaccharides to decide the extraction protocol. Sevage method was used to precipitated the protein from the crude polysaccharides solution. The polysaccharides were separated and purified by Sephadex G-75 column chromatography. The hypoglycemic effect of FTAP were investigated in alloxan-induced diabetic mice.【RESULTS】The extraction rate of the polysaccharides by boiling-water process was 15.5%, and the extraction rate of the polysaccharides by alkaline solution process was 14.9%.The method of boiling-water extraction polysaccharides was selected using extract FTAP. The results of Sephadex G-75 column chromatography showed purified FTAP were gained. After orally administrated FTAP 44mg/(kg•d) and 264mg/(kg•d) to hyperglycemic mice for 7d, the glucose and TG in serum were decreased obviously, but serum TC did not change remarkably, compared with the control group.【CONCLUSION】FTAP exhibited significant hypoglycemic activity in alloxan-induced diabetic mice.

The radio of raw material to water、extraction temperature and time were selected as three factors to design the orthogonal experament and the optimum condition of extraction techniques was obtained .The result showed that extraction temperature was the main factor .The optimum condition for the A rctium lappa extraction tecnique was as following :radio of raw material to water is 1:20,extraction temperature is 70℃,extraction time is 1 hour ; The optimum condition for the Giycine max(L)Merrill extraction tecnique was as following :radio of raw material to water is 1:10,extraction temperature is 70℃,extraction time is 1.5 hour ; The optimum condition for the Allium sativum extraction technique was as following :radio of raw material to water is 1:20,extraction temperature is 90℃,extraction time is 2 hour.

Methods A 722E spectrophotometer is analyse the anthra quinones was used to determine at different decocting time, while the time-content relation was analyzed. Results is the Rheum of ficinale decocted for 30min before, the content of free anthrax quinones increased rapidly reached the peak value at the end of 50min. Conclusion Rheum of ficinale and the dynamic changes of the decocting time. According to the experimental results, 50min is considered to the decocting time when Rheum of ficinale is used cathartic.

Meanwhile, by noting existing problems, some suggestions were proposed on strengthening risk assessment and safety management of GM trees in China.

  Study on Biomass and Productivity of Chinese Fir Plantation Hou Zhenhong, Zhang Xiaoquan, Xu Deying, Yu Pengtao 2009, 25 (5):  97-103.  Abstract + ( 1596 )   PDF ( 1284486KB )   ( 1001 ) Based on the Chinese Fir plantation biomass and productivity database, we analyzed the biomass, productivity, allocation rate of various organs in the Chinese Fir plantation, the relationship between the stand age and the biomass, productivity, allocation rate of various organs on the region scale. Results show that there is a significant relativity between the gross biomass of the Chinese Fir plantation and the stand age on the region scale. This relationship can be expressed by the Chapman-Richards growth equation. The biomass of every age calculated using the volume data in the forest inventory material is better than the biomass data measured in the experimental site in representing the actual growth situation of Chinese fir plantation. The biomass distribution rate of various organs is different with the stand age. The change trend of average productivity with the stand age is about ten years later than that of year-by-year productivity. So, the average productivity can not represent the yearly actual productivity change of the Chinese fir plantation. These results can provide some datum for the second national Greenhouse Gas Inventory and improve the quality of the Inventory.

  Influence of Pinus Massoniana on Soil Organic Matter Content of the Karst Rocky Desertification Area ，North-west of Guangxi Province Qin Yongrong,, Wang Yan, Liu Xuhui, Xu Jinqiang 2009, 25 (5):  104-109.  Abstract + ( 1453 )   PDF ( 813595KB )   ( 671 ) This paper is mainly on the influence of soil organic matter (SOM) content of Pinus massoniana forest during the process of ecological restoration in karst rocky desertification area, the northwest of Guangxi. The correlation between SOM of the plots and the ecological factor, such as aspect, altitude, recovery time, vegetation types were analyzed. It is showed that the SOM of Pinus massoniana was positively correlated to soil moisture, conductivity, amount of litter and cellulose decomposition strength, but negative correlated to the organic matter of litter. The influence of aspect on SOM is obviously, generally speaking, the soil organic matter content of the southeast slope is much higher than that of the northwest slope. In a certain range of time, the longer of the vegetation recover time, the higher of the soil organic matter content. Although, there is a downward trend of SOM in Pinus massoniana forest with the increase of the altitude. Because the ecological environment is very fragile, so, it is easy affected by human activity. Different frequency and different intensity of man-made interference results in different changes of SOM, the stronger of disturbance, the much lower of SOM, in the end, the soil will become more barren.

  Study on tissue culture and leaf regeneration of two kind of different provenances Siberian elm Wang Jinghua,Hou Jiansheng, Liu Guilin, Liang Haiyong, Yu Xiaoqing 2009, 25 (5):  110-115.  Abstract + ( 1218 )   PDF ( 1701067KB )   ( 628 ) Taking stem as the primary explants, a tissue culture system was established by using medium added with hormone at different concentration for two provenances of Siberian elm obtained from Cangzhou, Hebei; Chifeng, Neimeng. The results showed that the best differentiation medium for stem from Cangzhou is MS +6-BA 0.10㎎/L+IBA0.005㎎/L and for stem from Chifeng is MS+6-BA 0.10㎎/L+IBA0.025㎎/L; the best rooting medium for stem from Cangzhou is MS +IBA0.01㎎/L and for stem from Chifeng is MS +IBA0.05㎎/L. MS medium supplemented with 0.005mg/L TDZ and 0.005㎎/L IAA was the optimal medium condition of leaf regeneration of these two Siberian elms.

  Characteristics of Water Consumption by Transpiration of Zenia insignis at seedling age 2009, 25 (5):  116-120.  Abstract + ( 1182 )   PDF ( 1701068KB )   ( 891 ) Characteristics of water consumption by transpiration of Zenia insignis at seedling stage were studied by using pot seedling in different weather and water conditions.The results showed that the diumal variation of water consumption rate by transpiration of Zenia insignis showed single-peak curve.Water consumption rate of the current-year seedlings was higher than annual stubble seedlings.The largest water consumption rate of the current-year seedlings occured at 12:00—16:00 in sunny,but occured at 12:00—14:00 in cloudy.However,the annual stubble seedlings occured at 10:00—14:00 in sunny,occured at 10:00—12:00 in cloudy.Under water controlling,the permanent wilting coefficient of the current-year seedlings was 7.83%,annual stubble seedlings was 5.95%.When soil moisture was less than 6.03%,the current-year seedlings would die.When it was 3.32%,the annual stubble seedlings would languish permanently,even to die.

  Grey relational analysis of storage root yield and above-ground traits in different sweet potato genotypes 2009, 25 (5):  125-131.  Abstract + ( 1416 )   PDF ( 1537739KB )   ( 694 ) To study the correlation between yield of storage roots and above-ground agronomic traits, grey relational analysis was performed using sweet potato harvested from the Research Station at Tai’an city. Thirteen agronomic traits of sweet potato have been studied. Our results demonstrate that the five agronomic traits showing strong correlation with fresh weight of storage roots in order of importance are the fresh weight of branch petioles, the number of branch leaves, the total fresh weight of above-ground biomass, the fresh weight of branches and the number of branches. The top five traits significantly contributed to dry weight of storage roots are the fresh weight of branch leaves, the length of branches, the fresh weight of branch petioles, the number of branches and the number of branch leaves. Among the meteorological factors analyzed, the temperature was proved to be more important than humidity for a higher yield. Our study suggests that different traits need to be considered besides the major yield factors in order to reach a balanced objective in sweet potato breeding program.

So that hybrid rice panicle reached 3000000 / hm2 around, full grains reached 180-210 per panicle, spikelets reach 200-230 per panicle, stabilizing setting rate at 80-90 per cent, an effective solution to the contradictions of base ,source, flow with adequate panicle number and large panicles, and the realization of yield and quality objectives of 1200 kg / hm2.

  Analysis of agronomic characters of Xinjiang spring wheat 2009, 25 (5):  151-154.  Abstract + ( 1550 )   PDF ( 1463565KB )   ( 843 ) Agronomic characters play a important role in improving breed of wheat。The agronomic characters of 30 Xinjiang spring wheat were evaluated by using the statistical methods of simple and partial correlation analysis ,multiple stepwise regression and path analysis in order to provide helpful information for the genetic improvement of wheat1。We found that the plant height of most landraces was too high to be used for breeding program and the 1000-grain weight and grain weight per plant of them were lower, with the mean of 30.04 g and 8.84 g, respectively. 74% variation of grain weight per plant could be explained by the multiple regression equations Y = 0.6139 + 0.02X3 + 1.07 X4 +0.42 X5. So how to correspond every agronomic character should regard as important criteria of selecting in high yield breeding.

The results showed that compared with Zhengdan 958, Anlong 4 and Ludan 981 had bigger plants, higer leaf area index at every stage and longer functional period of leaf while the grain filling stage of that were prolonged 5-7 days. Meanwhile, single panicle weight of Anlong 4 and Ludan 981 were increased about 56.0-76.8g, and the grian weight of that were increased by 13.11～20.58%. Anlong 4 had obvious superiority of resistance to maize leaf spot, rust, and borer. Liyu 16 and Ludan 981 had better adaptability and disease resistance, and had signifitant resistance to maize borer. At the same time, Zhongke 11 had light sensitive to maize borer while Huadan 986 had high sensitive to that thus affect yields. So, under the ecoliogical environment condition of Taihe county, Anlong 4 and Ludan 981 had better comprihensive resistance and bigger yield potential, thus they coule be planted in large scale.

When the pollution caused by Zn at the low concentration of Zn (10mg•L-1)，the plant external injury of Alternanthera philoxeroides was not significant. At the concentrations of Zn (＜50mg•L-1)，the protection enzyme in cells such as SOD，POD and CAT increased. But with the Zn level increasing, the plants grew slowly, the biomass decreased, and the chlorophylls level decreased. Meanwhile the balance of these kinds of enzymes was destroyed in the active oxygen scavenging system。The accumulation of O2•- and H2O2 cannot be prevented effectively, the cell membrane permeability increased, the MDA content accumulated highly. Thus the growth of plant was affected, and toxic symptoms appeared.

The results showed that the biomass of cherry-radish affected by pig manure in the first harvest was better than that of wheat straw. Moreover, high rate usage of pig manure enhanced plant growth and the concentrations of nitrogen phosphorus and potassium in plant shoots. But wheat straw application lowered the dry weight of plant roots and the concentration of nitrogen in plants. The concentrations of total nitrogen and available potassium in soil were increased by the application of wheat straw. The contents of organic matter, total nitrogen, available phosphorus and available potassium in soil were increased by utilization of pig manure. The addition of pig manure resulted in a reduction of soil pH value, while a rise of soil EC level. The middle and high application rates of wheat straw also lead to an increase of soil EC. The present study suggested that wheat straw application should be combined with certain rates of nitrogen and phosphorus fertilizers to be adjusted a suitable C/N ratio. The available phosphorus concentration in soils would be enhanced by the addition of pig manure.

The result show that: different combined application all could improve the yield of alfalfa, among all the treatments the effect of single application Mo was the best, the follow was B and Mo combined application treatment, compared to CK, increase was 54.75%, 40.34%, respectively. Different B, Mn and Mo combined application has different influence on mineral element contents and accumulation amount of alfalfa. combined application micronutrient fertilizers decreased Fe, Mn and Zn contents, and B and Mo combined application has an obvious effect. But they could increase the Cu content of alfalfa, and the effect of B, Mn and Mo combined application was the best. B+Mn and B+Mo treatments could singificantly increaser B and Mo contents of alfalfa, respectively. Different combined application all could improve the mineral element accumulation amount of alfalfa. Single application of B could emarkably increase Fe, Mn and Zn accumulation amount, while single application of Mo and B, Mn and B, Mo combined application could significantly increase Cu, B and Mo accumulation amount of t alfalfa respectively.

The results showed that the biomasses of green Chinese onion in applied chelate fertilizer treatment were higher than those in conventional fertilizer treatment and control treatment during August, September and October, but during November there were no obvious difference on the yields between chelate fertilizer and conventional fertilizer treatments. The plant and welsh-onion stalk heights of chelate fertilizer treatment were higher than those of conventional treatment during the different harvest stages. Chelate fertilizer application could markedly enhance the nutritive quality of green Chinese onion than that of conventional fertilizer, it not only increased Vitamin C content but also deceased nitrate content in welsh-onion stalk. Thus, chelate fertilizer treatment was obviously better than conventional fertilizer treatment under the situation of the same fertilizer rates. In conclusion, altering conventional fertilizer kinds can improve the quality of green Chinese onion.

Shoot tips of Lysima chiachristinae Hance were used as explants to initiate buds on MS supplemented with 6-BA0.5,NAA0.02; subcultured on MS supplemented with 6-BA0.5, NAA0.05; roots were induced on 1/2MS supplemented with IBA0.5. The medium contained 30g /L sucrose and 6 g /L agar. The cultures were maintained at 25℃ under a 12-h photoperiod with light intensity 1800 Lx, can produce complete plants.

The aim of this study was to provide original data and information to instruct the chance seedling selection. The results indicated: ①The fruit weight and SSC content revealed rich genetic diversity in the sweet cherry naturally pollinated seedlings whose coefficients were 32.67% and 20.03%. However the coefficient of the edibility was lower (2.23%) which showed a narrower variation range. ②The contents of fructose, glucose and total sugar in the sweet cherry naturally pollinated seedlings also changed a lot. Their coefficients were 17.93%、17.05% and 17.40% respectively. And the coefficients of the contents of organic acid and sugar/acid value were larger, which were 25.67% and 35.22%. ③Ripe fruits of 31 sweet cherries were analyzed using head space-solid phase micro-extraction and gas chromatography-mass spectrometry. The results showed considerable genetic variations in these aspects: the number of volatile components category, the categories and contents of each kind of compounds and contents of main components. There were 113 compounds in total belonging to 5 classes detected, including esters, alcohols, ketones, aldehydes and hydrocarbons. In conclusion, the 31 sweet cherry seedlings showed wider variation range which indicated the possibility to obtain superior seedling.

  Pot Experiment of Allelopathy of Eupatorium catarium Veldkamp on Several Common Weeds in South China 2009, 25 (5):  250-253.  Abstract + ( 1787 )   PDF ( 481430KB )   ( 796 ) To explore the allelopathic effect of Eupatorium catarium Veldkamp as green manure on common weeds in the field, five plants(Eupatorium odoratum L., Mimosa pudica L., Echinochloa crusgalli (L.)Beauv, Bidens pilosa L. and Abutilon theophrasti Medic) were selected as receivers in pot experiment and the allelochemical donator was Eupatorium catarium Veldkamp. The rate of emergence, stem length, aboveground biomass and underground biomass of receiver plants were measured. The results indicated that: (1) Eupatorium catarium Veldkamp had strong inhibition to the germination of weeds, of which it was obvious on Echinochloa crusgalli (L.)Beauv and Eupatorium odoratum L.. The germination of both plants was totally inhibited under low application dose. (2) Relatively weak inhibitory action was shown to weed seedlings and even the growth of some plants was promoted. Therefore, our experiments confirmed that the time during which the weed could be effectively suppressed was before germination; meanwhile, the control object should be sensitive to allelopathy.

The plant demand little N, P, K at the earlier period, after the plant emerging, the total absorbed nutrient (TAN) was insufficient of 1/3 at the first of 70 days, then raised greatly until the harvest time, and the TAN was almost the 2/3 at the late of 40 days. The shell length (x1), the shell width (x2), the shell height (x3) and the live body weight (Y) were measured. The correlation coefficient matrix was calculated. The preceding 3 shell characters were used as independent variables, and live body weight was used as a dependent variable for path analysis, path coefficients, determination coefficients and correlation index were calculated, and major morphometric attributes determined. The results showed that all the correlation coefficient between each independent variable and dependent variable (body weight) were significantly different (P<0.05). The shell height gave a predominant direct effect ( 0.589 ) and determinacy on the body weight, it is the key effective factor, whereas shell length and shell width exhibited a slight direct effect (0.4023, 0.3787) on the body weight via shell height. The value of multiple vorrelation index R2 at 0.828 between morphometric and body weight suggests that the selected attributes are practical.The multiple regression equation relating the body weight was established as y=-265.233+0.743x1+2.459x2+3.688x3.

  Simple model for tomato and green pepper leaf area based on AutoCAD software 2009, 25 (5):  287-293.  Abstract + ( 1491 )   PDF ( 1408348KB )   ( 859 ) In order to get tomato and green pepper leaf area accurately and conveniently, scanner was used to get leaf images and AutoCAD was used to get actual leaves area. Regression analysis were carried on between actual leaf areas and leaf length, breadth, product of length-breadth measured in the field respectively, and regression models were established between leaf length, leaf breadth, product of leaf length-breadth and the actual leaves area respectively. The results showed that relations between the leaf length, the leaf breadth and the leaf area all assumes the power function, and relation between the product of leaf length-breadth and the leaf area assumes the linear. Errors analysis was carried on for three kinds of estimating models, and the results showed that accuracy of simulated value were bad for regression models of leaf length and leaf breadth. However, the estimated errors were much lower and precision was highest for the product of leaf length-breadth regression models for two kinds of crop, actual size of leaf area can be reflected truly, and the reduction coefficients were 0.6393 and 0.6509 respectively for tomato and the green pepper.

Throughout the history of water conservancy in modern China, he made great effort to inherit and develop traditional Chinese water control theory as well as to introduce and spread advanced water conservancy science and technology from western countries. He acknowledged the role of traditional Chinese levee, put the view of “academic embankment”; attached importance to the use of levee in river regulation; it functions as an extension of levee. He emphasized that the use of plants in dike consolidation and beach protection should be avoid its soil loosen demerit and carried forward its merit in silting process. Moreover, Li also hoped to improve Chinese levee construction and management in an all-round way, and strengthen its basic scientific research, so that there could be an integration levee construction which combines the use of flood prevention, river regulation, irrigation and transportation.
